Comparison of the performance of the H. perezi qPCR assay with purified target DNA, and DNA from sediment spiked with a surrogate for the parasite. Standard curves were generated from 10-fold serial dilutions of DNA corresponding to purified cloned plasmid pES103 DNA (symbol ♦ slope of −3.39, R2 = 0.917), DNA prepared with the MoBio kit, from E. coli xcontaining plasmid pES103 (symbol ▲, slope of −2.70, R2 = 0.979), and DNA prepared from sediment spiked with E. coli containing plasmid pES103 (symbol ■, slope of −3.22, R2 = 0.993).
